Sex-dependent histopathological differences in AAV9-treated Cln8mnd mice. No sex-dependent histopathological differences were observed when comparing ASM accumulation in AAV9-treated Cln8mnd males and females in the VPM/VPL (A). AAV9-treated Cln8mnd females exhibit greater ASM accumulation within the S1BF at 4 months of age (B). AAV9-treated Cln8mnd males evidenced enhanced SubC burden at 8 and 10 months of age within the VPM/VPL (C) and at 24 months of age within the S1BF (D). Greater astrocytosis (GFAP+) was observed in AAV9-treated Cln8mnd males at 10 months of age within the VPM/VPL (E), but not within the S1BF (F). No sex-dependent differences observed in microgliosis (CD68+) in the VPM/VPL (G) or S1BF (H). Two-way ANOVA with Fisher’s LSD post-hoc.
