Rescue of Tf accumulation by wild-type Rbsn-5. Cells from CB (A-D) or PA (E-H) were transfected with plasmids encoding RFP (A,C,E,G) or RFP-Rbsn5 (B,D,F,H). After 48 h, cells were exposed to Alexa-488-Transferrin for 5–30 min, washed briefly and fixed. Representative images of RFP (A,E) or RFP-Rbsn5 (B,F) and corresponding Tf images (C,G and D,H). Light intensity of Tf signal per cell was quantified (I). Plotted are means and SEM of 50–100 cells from 4 independent experiments. Statistical significance was calculated using 2-tailed paired Student t-test, where the means of each time point are paired as indicated.
